Background
MYSM1 (Myb-like, SWIRM and MPN domain-containing protein 1) is a metalloprotease deubiquitinase that removes monoubiquitin from histone H2A at K119, functioning as a transcriptional coactivator by counteracting Polycomb-mediated gene silencing, and also cleaves K63-linked polyubiquitin chains from innate immune signaling molecules such as TRAF3, TRAF6, RIP2 and STING to dampen TLR, NOD2 and cGAS-STING pathways; through these dual nuclear and cytosolic activities MYSM1 regulates hematopoiesis, lymphocyte development, stem cell quiescence, inflammatory responses and tumor suppression, and its germline mutations cause a rare bone marrow failure syndrome characterized by leukopenia, anemia and multi-organ developmental defects.
